Overview

[6,6-2H2]-glucose is a nonradioactive, stable isotope-labeled form of glucose used as a metabolic tracer in clinical research. It contains two deuterium atoms at the 6-position of the glucose molecule. When administered intravenously, it labels the blood glucose pool and is subsequently incorporated into the DNA of dividing cells, such as T cells, B cells, and NK cells, during de novo purine and pyrimidine synthesis. This incorporation allows for the quantification of cell proliferation and disappearance kinetics using mass spectrometry. It is primarily utilized in exploratory clinical studies, such as those conducted by the Academisch Medisch Centrum, to measure lymphocyte turnover and immune reconstitution kinetics in patients following hematopoietic stem cell transplantation (HSCT).
